AAV for Pancreas Research: AAV8 Serotype, insulin & PDX1 Promoters | WZBIO

1. Background

The pancreas is a long, narrow gland and the digestive and endocrine organ of vertebrates, combining both endocrine and exocrine digestive functions.

As an endocrine gland, it is composed of cell clusters of varying size — the islets of Langerhans. The islets consist mainly of four cell types: A cells, B cells, D cells and PP cells. Their principal role is to regulate blood glucose levels by secreting insulin, glucagon, somatostatin and pancreatic polypeptide.

As an exocrine gland, it is composed of acini and ducts and mainly performs a digestive function. The acini secrete pancreatic juice, and the ducts serve as the channels through which pancreatic juice is discharged. The pancreas can secrete pancreatic juice into the duodenum through the pancreatic duct; the juice contains bicarbonate, trypsinogen and lipase, and is able to break down the carbohydrates, proteins and fats in food passing from the stomach into the duodenum.

2. Choosing the Injection Route, Dose and Promoter

When using AAV vectors for pancreas research, the following points generally require attention:

  • A. Serotype: the AAV8 serotype is usually recommended for pancreas research.
  • B. Promoter: the insulin and PDX1 promoters show good pancreas specificity.
  • C. Recommended dose: approximately 1×10E12 vg per mouse.
